Gold-Catalyzed Intermolecular Alkyne Oxyarylation for C3 Functionalization of Benzothiophenes

چکیده

C3-selective C–C bond formation on benzothiophenes is challenging, and few direct functionalization methods are available. A gold-catalyzed reaction of alkynes with benzothiophene S-oxides provides regioselective entry into C3-alkylated the C7-alkylated isomer as minor product. This oxyarylation works alkyl aryl substituted unsubstituted benzothiophenes. Mechanistic studies identify that sulfoxide inhibits catalyst [DTBPAu(PhCN)]SbF6, which also degrades forms unreactive complex [(DTBP)2Au]SbF6.

The three-component coupling of terminal alkenes with arylboronic acids and oxygen nucleophiles is described. The reaction employs a binuclear gold(I) bromide as a catalyst and Selectfluor reagent as the stoichiometric oxidant.
